Clarinex (desloratadine); Clarinex.com:"CLARINEX® is an antihistamine — a type of medicine that blocks the action of histamine, a natural substance in your body that is released by your immune system during an allergic reaction. Antihistamines are often used to help relieve the symptoms of seasonal and year-round allergies — itchy, watery eyes; sneezing; and runny nose — caused by exposure to indoor allergens such as dust, mold, pet dander, and outdoor allergens caused by pollen from trees, grasses, or weeds. CLARINEX® is also approved to treat the itching and rash due to hives from unknown causes. CLARINEX® is available by prescription only. And unlike most over-the-counter and some prescription antihistamines, CLARINEX® is non-drowsy when taken at the recommended doses. In clinical studies for seasonal and year-round allergies, the most common side effects with CLARINEX® (desloratadine) Tablets 5 mg were similar to sugar pill, including sore throat, dry mouth and fatigue. In clinical studies for ongoing itching and rash from hives of unknown cause, the most common side effects with CLARINEX® (desloratadine) Tablets 5 mg were similar to sugar pill, including headache, nausea, and fatigue."

Clarinex (desloratadine); Centerwatch.com:"Clarinex, in an oral tablet formulation, was approved for the treatment of seasonal allergic rhinitis (SAR)in adults and children over 12 years of age in December 2001. In February 2002, Clarinex tablets were additionally approved for the treatment of perennial allergic rhinitis (PAR) and for symptomatic relief of chronic idiopathic urticaria (CIU) in adults and children over 12 years old. Clarinex is a once-daily non-sedating antihistamine that provides 24 hour relief. An estimated 45 million Americans are affected by allergies. Allergic rhinitis (AR) is the most common allergic disorder in the United States, affecting up to 30% of the population.SAR is considered a response to outdoor allergens, while PAR pertains to indoor allergens. AR symptoms impact daily functioning at work, school and during leisure time, resulting in millions of absentee days each year. Urticaria, known commonly as hives, affects an estimated 20% of Americans at some point in their lives. The condition results from a reaction to a substance, most commonly food, drugs or a topical agent. Urticaria is characterized by itchy, swollen welts on the skin."

Generically known as desloratadine, the antihistamine has also been approved in treating allergic rhinitis. Desloratadine does not have the side effects commonly associated with over the counter antihistamines, according to Schering-Plough."

Clarinex (desloratadine); FDA.gov:"Desloratadine is a longacting tricyclic histamine antagonist with selective H1-receptor histamine antagonist activity. Receptor binding data indicates that at a concentration of 2 – 3 ng/mL (7 nanomolar), desloratadine shows significant interaction with the human histamine H1-receptor. Desloratadine inhibited histamine release from human mast cells in vitro. Human histamine skin wheal studies following single and repeated 5 mg doses of desloratadine have shown that the drug exhibits an antihistaminic effect by 1 hour; this activity may persist for as long as 24 hours. There was no evidence of histamine-induced skin wheal tachyphylaxis within the desloratadine 5 mg group over the 28 day treatment period. The clinical relevance of histamine wheal skin testing is unknown."

******************************************************

Clarinex (desloratadine); AdvanceForNurses.com:"Clarinex® (desloratadine, Schering) is the latest prescription antihistamine to hit the market for the treatment of allergy symptoms. As you may have guessed by the name, it is the next generation of Claritin® (loratadine). Desloratadine is the most active metabolite of loratadine; indeed, it is 14-17 times more potent than its predecessor. Just like Claritin, Clarinex is non-sedating and well tolerated, with adverse effects similar to placebo. Available as a 5 mg tablet, it is dosed once daily without regard to meals and is indicated for patients ages 12 and older. It has yet to be approved for the pediatric population, and no dosage adjustment is required in the geriatric population. Clarinex is indicated for the treatment of seasonal allergic rhinitis and was studied both during spring and fall allergy seasons prior to approval. Once swallowed, it reaches peak plasma concentrations in 3 hours and has an elimination half-life of 27 hours. This is significant in that it results in true 24-hour coverage of allergy symptoms. Clarinex is equally eliminated via renal and fecal routes. Dosage adjustments are recommended for patients who are renally or hepatically impaired. These patients should take Clarinex every other day instead of daily. Other patient populations with specific concerns are pregnant women and nursing mothers. Clarinex received a pregnancy category C rating, and is known to pass through breast milk. Nursing mothers with significant allergy symptoms need to make a decision of discontinuing nursing or discontinuing Clarinex."
